چکیده

The role of Ca in transmitter release and facilitation has been examined at the squid giant synapse by evaluating the effect of changes in the external concentration of Ca on the excitatory postsynaptic potential (EPSP). Changes in Ca were achieved by means of an arterial perfusion technique, circumventing the diffusion barrier that exists between the synapse and the bathing medium. Transmitter release was found to follow a high power (n) of the external Ca at low nonsaturating Ca concentrations and low stimulus frequencies: 4.0 +/- 0.1 (mean +/- SE) in 22 experiments. The value of n was not fixed, however, but declined at stimulus frequencies above 0.3 Hz to 2.9 at Hz, 2.3 at 10 Hz, 1.7 at 50 Hz, and 1.1 at 80 Hz. This decline in n was due to a greater proportional facilitation of the EPSP at lower Ca levels. In a separate experiment, facilitation was determined at a fixed low Ca level over a wide range of stimulus frequencies. A plot of the maximum percentage increase in the EPSP against the stimulus frequency described three "steps," with plateaus occurring at 0.3-10 Hz, 10-50 Hz, and over 80 Hz. The timing of these steps corresponds to the stimulus frequencies at which the cooperativity declines. The results of this study indicate that the cooperative action of four Ca ions are required to trigger the transmitter release mechanism, and that the phenomenon of facilitation involves a reduction in Ca cooperativity. It is proposed that Ca ions can bind up to four receptors on the release site and that binding results in the opening of "gates." The opening of all four gates triggers transmitter release.(ABSTRACT TRUNCATED AT 250 WORDS)

Presynaptic calcium diffusion and the time courses of transmitter release and synaptic facilitation at the squid giant synapse.

At the squid giant synapse, a presynaptic action potential is accompanied by an influx of calcium ions that continue to be detectable with arsenazo III microspectrophotometry for several seconds. Nevertheless, transmitter release occurs phasically, lasting only about 2 msec. If a second action potential follows within about 100 msec after the first, it releases more transmitter. In this paper, ...

Processes Underlying One Porn of Synaptic Plasticity: Facilitation

Facilitation is one of the most prevalent forms of synaptic plasticity, and is often invoked as a quality which is important in the nervous system's ability to generate adaptive behavior. The squid giant synapse provides an excellent opportunity to explore the biophysical mechanism of synaptic facilitation. Previous studies showed that facilitation is not due to changes in presynaptic action po...

The calcium hypothesis and modulation of transmitter release by hyperpolarizing pulses.

Small presynaptic conditioning hyperpolarizing pulses reduce transmitter release to a depolarizing stimulus by a substantial amount, with little effect on release by a subsequent depolarization. This result, obtained at neuromuscular junctions and the squid giant synapse, has been offered as a disproof of the calcium hypothesis of transmitter release or the residual calcium hypothesis of synapt...

Transmission at voltage-clamped giant synapse of the squid: evidence for cooperativity of presynaptic calcium action.

Synaptic transmission was studied at the squid giant synapse with voltage clamp control of both presynaptic and postsynaptic cells. Because presynaptic voltage gradients can complicate interpretation of electrophysiological data obtained from this preparation, we used local Ca application to restrict Ca influx and transmitter release to a short and relatively isopotential portion of the elongat...

Facilitation of transmitter release at squid synapses

Facilitation is shown to decay as a compound exponential with two time constants (T1, T2) at both giant and non-giant synapses in squid stellate ganglia bathed in solutions having low extracellular calcium concentrations ([Ca++]o). Maximum values of facilitation (F1) were significantly larger, and T1 was significantly smaller in giant than non-giant synapses. Decreases in [Ca++]o or increases ...
